Optical tweezers for the study of protein structure, function and dynamics

Single molecule studies using the optical tweezers provide access to a broad range of important function-related information about proteins. This powerful technique has been used to study a many important biological processes such as protein folding, motor protein motion, enzyme function and degradation by proteases.
The latest developments in optical tweezers instrumentation allow the combination of single molecule force measurements with fluorescence, enabling the simultaneous readout of, for example, ligand binding events and the conformational changes that they induce. In addition to studying Hsp90 chaperone system, we plan to further push the capabilities of single molecule optical tweezers measurements combined with fluorescence by developing techniques which will enable us to study the conformational dynamics of membrane proteins such as transporters and translocases. I will outline the capabilities of this technique and our initial research plans, with the hope of inspiring discussion and possible future collaborations.
