Developing new dyes for bio-imaging: Chemistry to help you make the most of your microscope
New fluorophores with tailored (photo)physical, chemical, and biological properties are required to capitalise on rapid advances in optical microscopy. Using an example of our development of a series of boron-based fluorescent dyes for two-photon fluorescence microscopy, I will show how structure‒property relationships can be used to tailor optical performance of dyes (two-photon absorption cross-section, red-shifted absorption, photostability, quantum yield, and lifetime) and how further structural modification can be used to build on this understanding to produce dyes with the necessary properties for live-cell imaging. I will also briefly discuss some on-going projects in the development of dyes for gated-STED microscopy and in photosensitizers for photodynamic and photothermal therapy.
